Bioactivity TAT-CIRP is a a small peptide, refers to Trans-trans-activating (Tat)-cold-inducible RNA binding protein. TAT-CIRP is an inhibitor of myeloid differentiation protein 2 (MD2). TAT-CIRP exhibits robust neuroprotection against ischemic and hemorrhagic stroke in mice[1].
Invitro TAT-CIRP (5 μM) 通过 TLR4 非依赖性途径,减轻神经细胞中 NMDA 刺激(50 μM;1 小时)诱导的细胞凋亡和坏死[1]。
In Vivo TAT-CIRP (5-20mg/kg) 减轻大脑中动脉 (MCA) 闭塞 (MCAO) 小鼠缺血性中风后的脑损伤。至于 Tat,人类免疫缺陷病毒 1 型的细胞膜转导结构域,可用作对照肽[1]。TAT-CIRP (20 mg/kg;静脉注射;单剂量) 在小鼠中可通过血脑屏障,Cmax 值为 4762.6 μg/L,半衰期约为 90 分钟[1]。TAT-CIRP (20 mg/kg、100 mg/kg;静脉注射;每天一次,持续 7 天) 对小鼠几乎没有毒性,具有安全性[1]。TAT-CIRP 减少恒河猴的脑缺血损伤[1]。 Animal Model:
