Bioactivity Gol-NTR is a Golgi-targetable probe with high selectivity and sensitivity. Gol-NTR is Nitroreductase (NTR)-activated and has visualization acute lung injury (ALI) and repair function. Gol-NTR has a low detection limit of 54.8 ng/mL. Gol-NTR can be used for the research for monitoring and assessing treatment response of sepsis-induced ALI[1]. Storage: protect from light.
Invitro Gol-NTR 对 NTR 的荧光标记[1](1) 用 DMSO 溶液配制 1.0 mM Gol-NTR 原液。(2) 用 DMSO 溶液稀释原液配制 5.0 µM Gol-NTR 工作液。(3) 5.0 µM Gol-NTR 与 50 µM NADH 混合在含 5% DMSO 的 PBS 缓冲液 (10 mM,pH 7.4) 中,然后加入适量的 NTR。(4) 37℃ 孵育 30 min 后,在 405 nm (狭缝宽度:dex/dem=5/5 nm) 下记录光谱。Gol-NTR 对 A549 细胞中 NTR 的荧光标记[1](1) A549 细胞在不同氧浓度 (1%、5%、10%、15% 和 20% O2) 下培养 8 h。(2) 用磷酸盐缓冲盐水 (PBS) 冲洗 A549 细胞。(3) 5.0 µM Gol-NTR 处理 A549 细胞 1 h。(4) 用共聚焦荧光成像观察 A549 细胞的荧光图像。
In Vivo 体内成像[1](1) C57BL/6 雄性小鼠 (6-8 周龄,体重 20-22 g) 预注射 300 μL DMOG (25 mg/mL),24 h 后,腹腔注射 300 μL LPS (10 mg/kg),持续 6 h。(2) 颈椎脱臼处死小鼠,收集肺器官。(3) PBS 冲洗后,用 50 µM Gol-NTR 在 PBS 中孵育 1 h。(4) PBS 冲洗后,在小型动物成像系统上进行荧光成像 (420 nm 的激发波长和 510 nm 的发射波长)。
