Bioactivity | DRAQ5 is a novel cell permeant and far red-fluorescing DNA probe. DRAQ5 excites at a wavelength of 647 nm, close to the Ex, and produces a fluorescence spectrum extending from 665 nm out to beyond 780 nm wavelengths. DRAQ5 fluorescence reflects cellular DNA content. DRAQ5 can be used in combination with FITC and RPE-labelled antibodies, without the need for fluorescence compensation[1]. |
Invitro | 哺乳动物细胞全培养基染色方法[2]:(1)细胞铺板:消化分离细胞,在完全培养基中重悬至浓度为 2-4 × 105/ml。注:附着培养的细胞 (例如盖玻片或小室) 可以在原位染色, 1-2 ml染料孵育 4 cm2。(2)配制染色液:每 ml 培养基中加入 4 µl 5 mM 染料原液(终浓度为 20 µM)。注:核染色在 2.5-5 µM 即可检测,一般不超过 30 µM。(3)荧光染色:37℃ 孵育5-15 分钟。注意:不能出现过染现象。(4)清洗(选做): 37°C, 800 g,离心3- 5 分钟。弃上清,细胞以4 × 105/ml 重悬于含 10 mM HEPES (HY-D0857) 的完全培养基中。(5)流式细胞术:使用常规脉冲分析进行双重识别并使用适当的软件分析参数。(6)激光扫描显微镜:使用 695 nm 滤光片收集荧光图像。固定细胞染色方法[2]:(1)固定细胞:在 PBS 中使用 4% 多聚甲醛固定 30 分钟,在缓冲液 (如PBS) 中重悬。(2)荧光染色:使用与活细胞相似的染料浓度和相似的培养条件。 |
