Description XAP044 is a mGlu7-selective antagonist which inhibits lateral amygdala long term potentiation (LTP) in brain slices from wild type mice with a half-maximal blockade at 88 nm[1].
In vitro 先前的研究表明,与所有以往对mGlu7有选择性的化合物不同,XAP044的作用机制并非通过七次跨膜区域,而是通过位于mGlu7细胞外Venus flytrap领域的一个结合口袋作用,这一区域被认为是正交式激动剂的结合处,这一发现是通过重组细胞系中的嵌合体受体研究所提出的[1]。
In vivo 在啮齿类动物行为范式中,XAP044展现了良好的大脑渗透性以及广谱的抗压和类抗抑郁及抗焦虑效能。此外,XAP044能够减少巴甫洛夫恐惧行为获取期间的僵直反应和天生的焦虑,这与mGlu7缺失小鼠的表型一致。而且,XAP044能够在野生型小鼠的侧脑核切片中抑制长期增强(LTP),半最大抑制浓度为88 nm。同时发现,XAP044对mGlu7缺失小鼠的LTP无影响,表明这种药理学效应依赖于mGlu7 [1]。
