Description JNJ-42153605 is a mGlu2 receptor positive allosteric modulator (EC50: 17 nM).
In vitro JNJ-42153605对mGlu2受体的选择性进行了评估。研究发现,JNJ-42153605在30 μM的浓度下,对其他mGlu受体亚型既不表现出激动剂也不表现出拮抗剂活性。JNJ-42153605展现了高渗透性,且没有迹象表明存在P-糖蛋白质排出作用[1]。
In vivo JNJ-42153605通过抑制大鼠REM睡眠状态展示了中枢体内有效性,该效应在大鼠睡眠-觉醒EEG范式中以每千克体重3mg经口剂量观察到,此现象被证明是由mGlu2介导的。在小鼠中,JNJ-42153605以ED50为5.4 mg/kg皮下注射剂量,逆转了PCP引起的高活动量,暗示其具有抗精神病活性。JNJ-42153605从胃肠道的吸收速率快,0.5小时后达到最大浓度。其体内清除率在大鼠和狗中表现为中等至高(分别为35和29 mL/min/kg)。排除半衰期在不同物种中较短,大鼠为2.7小时,狗为0.8-1.1小时[1]。
