| In vivo | 在 8 次连续试验中,90 只卢万(LOU)大鼠和 91 只斯普拉格-道莱(SD)大鼠接受了小鸡 II 型胶原蛋白免疫,并服用了amiprilose HCl 水剂(1 mg/mL)。在 LOU 大鼠中,amiprilose HCl 组的关节炎发病率为 7/46(15%),而水组为 16/44(36%)(P 小于 0.01)。在 SD 大鼠中,实验组的发病率为 28/46(60%),对照组为 33/45(73%)(p 大于 NS),但实验组在第 16 天和第 21 天的关节炎发病率显著较低(p 小于 0.03)。在 LOU 实验中,amiprilose HCl 没有影响抗体滴度或对胶原的迟发型超敏反应,也没有影响 T 细胞亚群的分布。在 1 mg/mL的无毒浓度下,amiprilose HCl 可使培养的兔滑膜成纤维细胞中的 3H 胸苷掺入量降低 78%,并导致出现大量胞质内颗粒/空泡。amiprilose HCl系统还以剂量相关的方式降低了兔滑膜细胞上清液中前列腺素 E2 的水平,降幅高达 73%,但不影响胶原酶活性。[5] |
