Bioactivity Xylene Cyanol FF is an acid triphenylmethane dye. Xylene Cyanol FF can be used for histochemical staining of hemoglobin peroxidase or as a tracking dye for DNA sequencing in electrophoresis. Xylene Cyanol FF will be catalyzed by Fe and Al to accelerate oxidation under the addition of double oxidant hydrogen peroxide and potassium periodate. Xylene Cyanol FF thus enables the spectrophotometric determination of Fe and Al in the solution to be tested[1][2].
Invitro Xylene Cyanol FF 能够在不同聚丙烯酰胺凝胶电泳示踪 DNA。操作如下[1]:1.用于变性凝胶。(1)配制凝胶:10% 的丙烯酰胺 (19:1,丙烯酰胺:双丙烯酰胺),以及 8.3 M urea。使用条件为 55 ℃。(2)配制电泳缓冲液:89 mM Tris,HCl,pH 8.0,89 mM 硼酸,2 mM EDTA (TBE)。(3)配制上样缓冲液:10 mM NaOH,1 mM EDTA,以及 0.1% Xylene Cyanol FF。(4)凝胶在 IBI 型号 STS 45 电泳装置上以 70 W (50 V/cm,恒定功率) 运行,或在 Hoefer SE 600 电泳装置上以 60 °C (31 V/cm,恒定电压) 运行。(5)将凝胶置于 Whatman 3MM 纸上干燥并在 X 射线胶片上曝光。曝光时间长达 15 小时。2.用于变性凝胶。(1)配制凝胶:含有 8% 的丙烯酰胺 (19:1,丙烯酰胺:双丙烯酰胺)。(2)配制 DNA 悬液:含有 40 mM TrisβHCl,pH 8.0、20 mM 乙酸、2 mM EDTA 和 12.5 mM 乙酸镁 (TAEMg)。(3)煮沸 DNA 悬液并缓慢冷却至 16 °C。(4)配制染色液:含有 TAEMg、50% 甘油、0.02% 溴酚蓝和 0.02% Xylene Cyanol FF。用该染色液将样品调至 10 µL 的最终体积。(5)凝胶在 Hoefer SE-600 凝胶电泳装置上以 11 V/cm 的电压在 16 °C 下运行,并暴露于 X 射线胶片长达 15 小时或用 Stainsall (HY-D0987) 染料染色。
