Bioactivity W4022 (compound 16) is a highly potent lysine methyltransferases G9a/NSD2 dual inhibitor with IC50s of 0.241 μM and 0.017 μM for G9a and NSD2, respectively. W4022 shows potent antiproliferative activity and exhibits potency in inhibiting colony formation, inducing cell apoptosis, and blocking cancer cell metastasis. W4022 effectively inhibits the catalytic functions of both G9a and NSD2 in cells and exhibits significant antitumor efficacy in the PANC-1 xenograft[1].
