Bioactivity SAPE (1-Stearoyl-2-arachidonoyl-sn-glycero-3-phosphorylethanolamine) is an R-Phycoerythrin (HY-D0988) labeled Streptavidin (HY-P3152) fluorescent probe. SAPE can be used for tumor detection when combined with biotin. SAPE has high sensitivity and a wide detection range[1].
Invitro SAPE 用于肿瘤检测[1](1) 将偶联了肿瘤伴随诊断因子捕获抗体的微球超声振荡 20 s。(2) 按 50 个微球/µL 的终浓度将微球存储在检测缓冲液中。取 50 µL 到 96 孔板的孔中,随后加入 50 µL 检测缓冲液、50 µL 标准品作为对照。(3) 800 rpm/min 室温孵育 30 min。(4) 在磁性分离板上放置 30‑60 s,移去上清液并清洗两次 (每次 100 μL 反应缓冲液)。(5) 加入 50 µL 检测缓冲液,混匀。(6) 用检测缓冲液稀释生物素化的检测抗体至 4 µg/mL,每孔各加入 50 µL。(7) 800 rpm/min 室温孵育 30 min。(8) 在磁性分离板上放置 30‑60 s,移去上清液并清洗两次 (每次 100 μL 反应缓冲液)。(9) 加入 50 µL 检测缓冲液,混匀。(10) 用检测缓冲液稀释 SAPE 至 4 µg/mL,每孔各加入 50 µL。(11) 800 rpm/min 室温孵育 30 min。(12) 在磁性分离板上放置 30‑60 s,移去上清液并清洗两次 (每次 100 μL 反应缓冲液)。(13) 加入 100 µL 检测缓冲液,混匀。(14) 激发 Luminex 磁性微球的荧光编码信号和 SAPE 的报告荧光信号,再通过高灵敏度的 CCD 成像装置或流式分析装置并由软件经特定的算法进行 Luminex 磁性微球分类及结果检测。
