Bioactivity Fluo-3FF AM is a low affinity (Kd = 42 μM) fluorescent Ca2+ sensitive indicator (Abs/Em = 462 nm/526 nm). Fluo-3FF AM is Mg2+ insensitive and relatively photostable. Fluo-3FF AM is an analog of Fluo-3FF AM. Fluo-3FF AM is essentially non-fluorescent, but exhibits a strong fluorescence enhancement upon entry into cells and binding to calcium.[1]
Invitro 1. Fluo-3FF AM 工作液的配制1.1 制备储存液用0.025% (w/v) 的 Pluronic F-127/二甲基亚砜溶液稀释 Fluo-3FF AM 去配制 1 mM 的储备液。注:Fluo-3FF AM 储存液建议分装后于 -20℃ 或 -80℃ 避光保存。1.2 工作液的配制用平衡缓冲溶液,配制成 5 μM 的 Fluo-3 AM 工作液。注:请根据实际情况调整 Fluo-3FF AM 工作液浓度,且现用现配。2.细胞染色2.1 将贴壁细胞培养于无菌盖玻片上。2.2 从培养基中移出盖玻片,吸除多余培养基。2.3 加入 100 μL 染料工作液,轻轻晃动使其完全覆盖细胞,孵育 60 分钟。2.4 吸去染料工作液,然后在 4◦C 的 PSS 中清洗 60 分钟。
