Bioactivity Chlorpyrifos is a neurotoxic insecticide that belongs to the class of thionite esters. Chlorpyrifos is also a AChE inhibitor that affects neurological function in insects, humans and other animals. Chlorpyrifos interferes with cell replication and differentiation, ultimately altering synaptic transmission in neurons[1][2][3][4].
Invitro 毒死蜱通过调节脱硫产生毒死蜱氧酮 (CPO), CPO 对丝氨酸依赖性酯水解酶的活性位点 (如AChE) 具有较高的亲和力[2]。毒死蜱 (3.9-250 μM;24-72 h) 对少突胶质细胞祖细胞具有细胞毒性[3]。毒死蜱 (7.5-480 μM;18 h) 导致细胞核凝结,caspase 3/7 活性升高,(60 μM; 2, 4 h) 还增加中枢胶质细胞血红素氧合酶-1 mRNA 表达,(30, 60, 120 μM;24 h) 增强 H2DCF-DA 强度[3]。 Cell Viability Assay[3] Cell Line:
In Vivo 毒死蜱 (97-276 mg/kg; p.o.; single dose) 有中度急性口服毒性,对大鼠的 50% 的致死剂量 (LD50) 为 97-276 mg/kg[2]。毒死蜱 (1 mg/kg and 5 mg/kg for 1 mL/kg; s.c.; once daily for 3 d) 对大鼠的学习和记忆能力有不良影响,在妊娠 9-12 天处理妊娠大鼠,导致子代行为异常[4]。
