Bioactivity | CAA-0225 is a potent inhibitor of cathepsin L, showing significant specificity towards this enzyme over cathepsin B. It inhibits rat liver cathepsin L with an IC50 value of 1.9 nM, while exhibiting minimal inhibition of cathepsin B. In studies on autophagy, CAA-0225 effectively suppresses the degradation of long-lived proteins under nutrient-deprived conditions in HeLa and Huh-7 cells. It also inhibits the degradation of autophagosomal membrane markers, such as LC3-II and GABARAP-II, indicating its role in blocking the degradation process specifically associated with these markers, rather than general lysosomal protein degradation[1]. |
