| Invitro | Guidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).BODIPY FL Thapsigargin[1]:1. Purified parasites are fixed with 1% formaldehyde in PBS (pH 8) for 1 h at room temperature and allows to adhere to poly-L-lysine-coated glass slides for 20 min Parasites are then permeated with 0.1% Triton X-100 for 5 min and blocked with 3% bovine serum albumin in PBS for 1 h.2. Parasites were incubated at room temperature with a 1:100 dilution of the primary polyclonal antibody H-300 for 1 h, followed by a 1:100 dilution of Texa Red-coupled goat anti-rabbit IgG secondary antibody, and with 0.25 µM BODIPY FL TG for 5 min.3. Analyze sample on a flow cytometer, fluorescence microscopy, or fluorescence microplate reader. |
