Bioactivity

Ac-DEVD-AMC is a fluorescent substrate of caspase-3/caspase-7. When treating Ac-DEVD-AMC with cell lysate, Ac-DEVD-AMC releases amino-4-methylcoumarin (AMC) for fluorescence detection, with an excitation wavelength of 380 nm and an emission wavelength of 460 nm[1][2][3].

Invitro

Ac-DEVD-AMC 荧光标记[2](1) 制备 caspase 缓冲液：0.5% Igepal CA-630、10 mM HEPES (pH 7.4)、2 mM EDTA、0.5 mM 苯甲基磺酰氟 (PMSF)、5 µg/mL 亮抑酶肽。(2) 50 µM 6-OHDA 处理 PC12 细胞 20 h。(3) 250 g、5 min 离心收集细胞，PBS 洗涤后在 250 g、5 min 下离心。(4) 按 2 µL/105 个细胞的比例，用 caspase 缓冲液重悬细胞，4°C 下裂解 20 min。(5) 7200 g、4°C、10 min 离心收集裂解物。(6) 20 µL 裂解物与 100 µM Ac-DEVD-AMC 在 30°C 下共孵育 10 min。(7) 在激发波长为 380 nm、发射波长为 460 nm 下用荧光分光光度计测量 caspase 活性。
