Invitro	AF488 NHS ester 的 IgG 荧光标记与抗 IgG 固相肽库筛选[2](1) 将冻干的 IgG 以 5 g/L 的浓度溶解在 50 mM 磷酸钠、20 mM 氯化钠、PH 8.3 的溶液中。(2) 1 mg AF488 NHS ester 溶于 100 µL extra dry DMF 中，加入 1 mL (1) 中溶液，室温旋转孵育 1 h。(3) 用 Amicon Ultra 0.5-mL 离心式过滤装置，配有 3-kDa MWCO 过滤器收集样品。(4) 六聚体或四聚体去保护文库在 50 mM 磷酸钠、150 mM 氯化钠、PH 7.4 (PBS) 溶液中以 5× 沉淀树脂体积洗涤三次以平衡。(5) 用 50 mM 磷酸钠、150 mM 氯化钠、0.2% 吐温 (PH 7.4) 溶液稀释 IgG-AF488 至终浓度为 1.3 mg/mL。(6) 将 (4) 和 (5) 在 2-8°C 下孵育过夜。(7) 用 50 mM 磷酸钠、150 mM 氯化钠、0.1% 吐温 20、PH 7.4 (PBS-T) 洗涤树脂珠。(8) 将树脂在 40 µL PBS-T 的 96 孔板中每孔沉积 1 个珠，然后使用荧光显微镜以 10 倍放大率成像。在 480 nm 激发下，以 510 nm 发射强度为阈值进行 Alexa Fluor 488 荧光测量和荧光筛选。AF488 NHS ester 储存液[1]用 DMF 配制 20 mM 的 AF488 NHS ester。注：AF488 NHS ester 储存液建议分装后于 -20°C 避光保存。
