| ln Vitro | An essential step in cellular metabolism is phosphoribosyl pyrophosphate (PRPP) pentasodium. The following is the process by which PRPP synthase makes PRPP: ribose 5-phosphate + ATP → PRPP + AMP. Living things contain PRPP in large quantities. It is utilized in substitution processes to create glycosidic linkages. The amino acids histidine and tryptophan, the cofactors NAD and tetrahydromethotrexate, arabinose monophosphate, and some aminoglycoside antibiotics are all biosynthesised via PRPP [1]. In human erythrocytes without hypoxanthine-guanine phosphoribosyltransferase accumulation, phosphoribosyl diphosphate (5-Phosphoribosyl-l-pyrophosphate) pentasodium is a substrate shared by adenine phosphoribosyltransferase and hypoxanthine-guanine phosphoribosyltransferase. Purified adenine phosphoribosyltransferase is stabilized against thermal inactivation by adding 5-phosphoribosyl-l-pyrophosphate [2]. |
