Biological Activity


ln Vitro Guide (the following is our proposed plan, this plan merely gives a guide and can be modified according to your individual needs). Binding of SELP analogues to Cy5-SE [1]. 1. Take 1 mL of 100 mM sodium bicarbonate buffer. 2. Quench reaction: mix 50 μL of 100 μg/mL SELP solution made of 1 M (pH 8.3) and 10 μL of 1.2 mg/mL Cy5 monomer composed of 10% DMSO. NHS ester was combined and kept on ice for 2 hours. 3. Load the reaction mixture onto a 1.5 mL Sephadex G-25 column, and centrifuge at 1050 g for 3 minutes to elute Cy5-labeled SELP. Note: 1. CY dye will not significantly interfere with peptides 2. Low amounts of sodium azide (≤3 mM or 0.02%) or thimerosal (≤0.02 mM or 0.01%). 3. Avoid utilizing buffers containing primary amines (such as Tris, glycine) or ammonium ions. They will compete with the protein to be tagged and impair labeling efficiency. 4. For dyes without activator, you need to use condensation solution (500 ug/mL) (25952-53-8) activator candle for future marking tests. 5. This product is solely for scientific research by professionals and may not be used for clinical diagnosis or treatment. 6. For your safety and health, please don a lab coat and wear a disposable overview for operation.

Solubility (In Vivo) Solubility in Formulation 1: ≥ 2.5 mg/mL (3.32 mM) (saturation unknown) in 10% DMSO + 40% PEG300 + 5% Tween80 + 45% Saline (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 400 μL PEG300 and mix evenly; then add 50 μL Tween-80 to the above solution and mix evenly; then add 450 μL normal saline to adjust the volume to 1 mL.
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.

Solubility in Formulation 2: ≥ 2.5 mg/mL (3.32 mM) (saturation unknown) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), clear solution.
For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution.

Solubility in Formulation 3: 25 mg/mL (33.16 mM) in Saline (add these co-solvents sequentially from left to right, and one by one), clear solution; with ultrasonication (<60°C).
Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH₂ O to obtain a clear solution.
