Description L 012 sodium salt, a luminol analogue, is a widely used chemiluminescent (CL) probe for active oxygen and active nitrogen. L 012 sodium salt is widely used to detect the superoxides (O2•−) derived from NADPH oxidase (Nox) to find Nox inhibitors.
In vitro L 012 sodium salt是一种新型化学发光探针。在产生活性氧物种(ROS)的激活EoL-1细胞中,L-012的处理产生了明显的化学发光。而超氧化物歧化酶(100-300 U/ml)消除了化学发光,这表明主要检测到的ROS是O2- [1]。在培养的内皮细胞中,佛波醇12-肉豆蔻酸13-酯(PMA,1 μM)使L-012的化学发光增加了2.8倍。钒酸盐(100 μM)将化学发光增加至17倍。这些结果表明,由NAD(P)H氧化酶产生的超氧增加。
In vivo 在人体口腔、血液以及大鼠腹膜腔中,L-012与活化中性粒细胞产生的ROS发生反应,产生了强烈的化学发光,其亮度高于MCLA [2]。在非肥胖糖尿病小鼠和K8敲除(K8(-/-))小鼠中,通过给予硫酸右旋糖醇的处理,增加了L-012的化学发光,用于检测反应性氧和氮种类(ROS和RNS)[3]。
