| In vitro | 方法: Dihydroethidium 检测细胞过氧化物:1、用 DMSO 配制 5-10 mM 的 Dihydroethidium 染料储备液,避光,分装储存于 -20℃。2、使用时用生理缓冲液 (如 PBS、HBSS、HEPES) 稀释储备液,得到 5-20 µM Dihydroethidium 染料工作液。最佳工作浓度必须根据经验确定。3、将染料工作液按 1:1 的比例加入到细胞中 (100 µL 细胞悬液加入 100 µL 染料工作液),在室温或 37℃ 下孵育 5-60 分钟。4、用 PBS 洗涤后,进行荧光显微镜或流式细胞仪检测。Dihydroethidium 本身为蓝色荧光,最大激发波长为 370 nm,最大发射波长为 420 nm,脱氢后与 RNA 或 DNA 结合产生红色荧光,最大激发波长为 300 nm,最大发射波长为 610 nm,实际观察时也可以使用 518 nm 作为激发波长。 |
