| In vivo | 方法:fluorescent microscope 检测循环乳腺癌细胞的葡萄糖摄取:1、通过穿刺小鼠隐静脉收集小鼠血液样本 (100 µL/mouse)。2、将含有循环乳腺癌细胞的血液样品与 2-NBDG (5 µg/100 µL 血液) 在 37℃ 的黑暗培养箱中孵育 30 min。3、将磁珠悬浮液 (1µL 1% )添加到 100 µL 的血液样品中,然后在 4℃ 下孵育 30 min,轻轻摇晃,以促进磁珠与循循环乳腺癌细胞结合。4、用磁性分离架分离从血液中收集循环的乳腺癌症细胞,用 PBS 洗涤 3 次,再次悬浮在 100 µL PBS 中后转移到 96 孔细胞板中。5、在配备 488 nm 滤光片的荧光显微镜下检查循环乳腺癌细胞对 2-NBDG 的摄取。具有来源于荧光 2-NBDG 的细胞摄取的荧光信号的大细胞被计数为高代谢循环乳腺癌症细胞,小尺寸的正常小鼠血细胞 (淋巴细胞和 RBC) 没有或很少显示 2-NBDG 荧光信号。[2] |
