| Cell experiments | Cells were seeded in 96-well plates at a density of 1*10^4 cells in a final volume of 100 μl and maintained in DMEM medium at 37℃ ?.?After cells were treated with different concentrations of HA (5, 10, 20, 40, 80 μM), 20 μl 5 mg/ml MTT solution were added to each well and incubated for 4 h in the incubator.?Then 150 μl DMSO were added to each well to dissolve the pellet, keep it in the dark for 15 min, and dissolve the crystal completely.?Automatic microplate reader measured at 490 nm optical density (OD value).?The experiment was repeated three times.?For combination test, cells were treated with 20 μM Hypocrellin A?and different?concentrations of OA(25,50,100, 200 μM) . |
