Bioactivity MQAE is a novel chloride ion fluorescence probe excited by ultraviolet light. MQAE, binding bromine ion as ligand, can be used to detect the activity of chloride. MQAE exhibits a reduction of fluorescence intensity with the increase of intracellular Cl- concentration. MQAE has a pH-independent fluorescence intensity basically at a Cl- concentration <50 nM. MQAE has high water solubility and cell permeability, MQAE has been used for fluorescence detection such as confocal microscopy and flow cytometry. Ex/Em=350/460 nm[1]. Storage: protect from light.
Invitro 1.MQAE 工作液配制1.1 制备储存液将 1 mg MQAE 用 0.3066 mL DMSO 配制 10 mM 储备液。1.2 工作液的配制使用 Krebs-HEPES 缓冲液 (20 mM HEPES,128 mM NaCl,2.5 mM KCl,2.7 mM CaCl2,1 mM MgCl2,16 mM glucose,pH 7.4) 配制 5-10 μM 的 MQAE 工作液。注:请根据实际情况调整 MQAE 工作液浓度,且现用现配。2 细胞染色2.1 准备工作悬浮细胞:离心收集细胞,加入 PBS 洗涤两次,每次 5 分钟。贴壁细胞:弃去培养基,加入胰蛋白酶消化细胞。离心弃去上清后,加入 PBS 洗涤两次,每次 5 分钟。2.2 加入 1 mL MQAE 工作液,室温孵育 30 分钟。2.3 400 g,4℃ 离心 3-4 分钟,弃去上清。2.4 加入 PBS 洗涤细胞两次,每次 5 分钟。2.5 用 1 mL 无血清培养基或 PBS 重悬细胞后,使用荧光显微镜进行观察。
