Description | Lys(Dabsyl)-(Asn670,Leu671)-Amyloid β/A4 Protein Precursor770 (667-676)-Gln-Lucifer Yellow also called Lys(Dabsyl)-(Asn670,Leu671)-APP770 (667-676)-Gln-Lucifer Yellow, is a highly selective substrate for measuring BACE1 (Km = 9 µM, kcat = 0.02 s-1) and BACE2 activity. In this fluorescence resonance energy transfer (FRET) substrate the fluorescent group Lucifer Yellow is efficiently quenched by Dabsyl (4-(4-Dimethylaminophenylazo)benzenesulfonyl). Enzymatic cleavage can be assayed by detecting the increase in fluorescence emission at 520 nm using an excitation wavelength of 430 nm. |
