Biological Activity
| ln Vitro | Beta-Amyloid Aggregation Guidelines: The protocol we recommend is as follows. You should adjust this process to suit your unique needs as it simply offers guidelines. Dissolve the solid Aβ peptide in cold HFIP (hexafluoro-2-propanol). To ensure that the structure is randomized and monomerized, incubate the peptides for a minimum of one hour at room temperature. 2. Put the peptide that results from evaporating to preserve it on a film at -20 or -80°C. This will eliminate HFIP. 3. After achieving the desired concentration and buffer (serum- and phenol-red-free media), dissolve the resultant membrane in anhydrous DMSO at a concentration of 5 mM, vortex, and dilute. The solution should then be aged for 48 hours at 4–8°C. The supernatant contained soluble oligomers after the samples were centrifuged for 10 minutes at 4–8°C at 14,000 g. 10–200 times washed supernatant was used in studies. The techniques used differ based on the final use. |

Solubility in Formulation 1: 2.5 mg/mL (3.36 mM) in 10% DMSO + 90% (20% SBE-β-CD in Saline) (add these co-solvents sequentially from left to right, and one by one), suspension solution; with sonication. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of 20% SBE-β-CD physiological saline solution and mix evenly. Preparation of 20% SBE-β-CD in Saline (4°C,1 week): Dissolve 2 g SBE-β-CD in 10 mL saline to obtain a clear solution. Solubility in Formulation 2: ≥ 2.5 mg/mL (3.36 mM) (saturation unknown) in 10% DMSO + 90% Corn Oil (add these co-solvents sequentially from left to right, and one by one), clear solution. For example, if 1 mL of working solution is to be prepared, you can add 100 μL of 25.0 mg/mL clear DMSO stock solution to 900 μL of corn oil and mix evenly.  (Please use freshly prepared in vivo formulations for optimal results.) |
