RdRP-IN-9 (Compound 6b) is a reversible covalent RdRp allosteric inhibitor. RdRP-IN-9 exhibits high anti-coronavirus activity, with an EC50 value of 0.68 μM against HCoV-OC43. RdRP-IN-9 can reversibly acylate Cys12 of the NiRAN domain in nonstructural protein 12 (nsp 12), thereby exerting a more persistent antiviral effect, and exhibits synergistic anti-coronavirus activity with Molnupiravir.
