Biological Activity


ln Vitro PNU-288034 is mediated via human multidrug and toxin extrusion protein 1 (hMATE1; Km = 340 +/- 55) uptake micron and human organic anion transporter 3 (OAT3; Km = 44 +/- 5 μM), according to in vitro investigations done to characterize key transporters[1].
ln Vivo PNU-288034 is metabolically stable in hepatic microsomes from all species, and in rats and monkeys, an apparent active renal secretory mechanism excretes the drug in urine unaltered. When PNU-288034 and the OAT3 inhibitor probenecid were given together, the plasma area under the curve for PNU-288034 increased dramatically (170%), and the plasma and renal clearance in monkeys were lowered. Rats' plasma clearance was likewise decreased by PNU-288034 and cimetidine coadministration to values similar to those of probenecid coadministration [1].

Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
