PF-06737007 (PF06737007) is a novel, potent, selective, peripherally restricted, efficacious, and well-tolerated pan-Trk inhibitor with anti-hyperalgesic activity. In cell-based assays, it inhibits TRK with IC50s of 7.7 nM, 15 nM and 3.9 nM for TrkA, TrkB and TrkC, respectively. It has anti-hyperalgesic effect. Hormones of the neurotrophin family, nerve growth factor (NGF), brain derived neurotrophic factor (BDNF), neurotrophin 3 (NT3), and neurotrophin 4 (NT4), are known to activate the family of Tropomyosin receptor kinases (TrkA, TrkB, and TrkC). Moreover, inhibition of the TrkA kinase pathway in pain has been clinically validated by the NGF antibody tanezumab, leading to significant interest in the development of small molecule inhibitors of TrkA. Furthermore, Trk inhibitors having an acceptable safety profile will require minimal brain availability.

Biological Activity


ln Vitro PF-06737007 (compound 13b) demonstrates superior selectivity for Trks, as seen by its over 95% inhibition of TrkA at a dose of 1 μM, while other kinases only show a 40% inhibition rate [1].

Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
