IACS-9571 (ASIS-P040) HCl is an inhibitor (blocker/antagonist) of bromodomain proteins TRIM24 and BRPF1. The IC50 and Kds for TRIM24 are 8 nM and 31 nM respectively, and the Kd for BRPF1 is 14 nM.

Biological Activity


Targets IC50: 8 nM (TRIM24)[1] Kd: 31 nM (TRIM24), 14 nM (BRPF1)[1]
ln Vitro IACS-9571 hydrochloride hydrochloride has an EC50 of 50 nM, which indicates good cellular potency. Strongly active against a group of 32 bromodomains is IACS-9571 hydrochloride (1 μM). With a selectivity of 9 and 21 fold for BRPF2 and BRPF3, respectively, IACS-9571 hydrochloride is a dual TRIM24/BRPF1 inhibitor that is selective (Kd=1.3/2.1 nM). With respect to BRD4, IACS-9571 hydrochloride hydrochloride exhibits about 7700-fold selectivity over TRIM24 and does not interact with the BET subfamily of bromodomains (1, 2) [1].

Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
Oral Formulation 3: Dissolved in PEG400
Oral Formulation 4: Suspend in 0.2% Carboxymethyl cellulose
