Hydrangenol is an orally bioavailable anti-photoaging compound. Can be extracted from the leaves of hydrangea (Hydrangea serrata). Hydrangenol prevents wrinkle formation by reducing the expression of MMPs and inflammatory cytokines and increasing the levels of moisturizing factors and antioxidant genes.

Biological Activity


Targets MMP-1 MMP-3 COX-2 IL-6
ln Vivo In UVB-exposed HR-1 hairless mice, hydrangeenol (5, 10, 20, or 40 mg/kg; oral; daily for 7 weeks) decreases the growth of wrinkles, back thickness, dehydration, and collagen breakdown[1].
Animal Protocol Animal/Disease Models: UVB-irradiated HR-1 hairless mice[1]
Doses: 5, 10, 20, or 40 mg/kg
Route of Administration: Oral gavage; daily for 7 weeks
Experimental Results: Increased the expression of involucrin, filaggrin, and aquaporin-3 (AQP3) as well as hyaluronic acid (HA) production via hyaluronidase (HYAL)-1/-2 downregulation. Increased the expression of Pro-COL1A1. diminished the expression matrix metalloproteinase (MMP)-1/-3, cyclooxygenase-2 ( COX-2), and interleukin-6 (IL-6). Attenuated the phosphorylation of mitogen-activated protein kinases (MAPKs) including ERK and p38, activator protein 1 (AP-1) subunit, and signal transduction and activation of transcription 1 (STAT1). Upregulated the expression of nuclear factor-E2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), NAD(P)H quinone dehydrogenase 1 (NQO-1), glutamate cysteine ligase modifier subunit ( GCLM), and glutamate cysteine ligase catalysis subunit (GCLC).

Additional Infomation Hydrangenol is a member of the class of dihydroisocoumarins that is 3,4-dihydroisocoumarin substituted by a hydroxy group at position 8 and a 4-hydroxyphenyl group at position 3. It has been isolated from the roots of Scorzonera judaica and exhibits anti-allergic activity. It has a role as an anti-allergic agent and a plant metabolite. It is a member of phenols and a member of dihydroisocoumarins. It is functionally related to a 3,4-dihydroisocoumarin.
Hydrangenol has been reported in Hydrangea serrata, Hydrangea macrophylla, and Hydrangea febrifuga with data available.

Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO 900 μL Corn oil)
Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals).

Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
Oral Formulation 3: Dissolved in PEG400
Oral Formulation 4: Suspend in 0.2% Carboxymethyl cellulose
Oral Formulation 5: Dissolve in 0.25% Tween 80 and 0.5% Carboxymethyl cellulose
