Aprinocarsen (ISIS 3521) sodium is a specific antisense oligonucleotide inhibitor of protein kinase C-α (PKC-α). Aprinocarsen sodium is a 20-mer oligonucleotide that regulates cell differentiation and proliferation. Aprinocarsen sodium suppresses the growth of human tumor cell lines in nude mice. Aprinocarsen sodium has value as a chemotherapeutic compound for human cancers.

Biological Activity


ln Vitro In human bladder cancer (T-24) cells, aprinocarsen sodium inhibits PKC-alpha in a dose-dependent manner and with a specific oligonucleotide sequence[1]. The PKC family of genes' other members, PKC-eta and zeta, are not affected by aprinocarsen sodium, however it has an IC50 value of 50–100 nM for PKC-alpha mRNA reduction[1].
ln Vivo Aprinocarsen sodium (100 mg/kg; intravenously; at intervals of dosing for 14 days) exhibits a well-tolerated profile with no discernible acute toxicity[1]. In nude mice, aprinocarsen sodium (iv) has dose-dependent growth inhibitory effects on human tumor cell lines T-24 bladder, human lung carcinoma (A549), and Colo 205 colon cancer, with ID50 values ranging from 0.06 to 0.6 mg/kg per day[1].

Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
