Aloisine A (RP107) is a potent cyclin-dependent kinase (CDK) inhibitor (antagonist) with IC50s of 0.15 μM and 0.12 μM for CDK1/cyclin B, CDK2/cyclin A, CDK2/cyclin E, and CDK5/p35 respectively. , 0.4 μM, 0.16 μM. Aloisine A inhibits GSK-3α (IC50=0.5 μM) and GSK-3β (IC50=1.5 μM). Aloisine A stimulates wild-type CFTR and mutant CFTR with submicromolar affinity through a cAMP-independent mechanism. Aloisine A may be used for studying CFTR-related diseases like cystic fibrosis.

Biological Activity


ln Vitro Erk1 (IC50=18 µM), erk2 (IC50=22 µM), and c-Jun N-terminal kinase (JNK; IC50~3-10 µM) are all inhibited by aloisine A. Protein kinase C α, β1, β2, γ, δ, ε, η, and Σ (all with IC50 values more than 100 µM) are unaffected by aloisine A [1]. Aloisine A (0.1, 1, 10, 100 µM) almost entirely prevents cell death while totally blocking the proliferation of differentiated postmitotic neurons (hNT; IC50=10.5 µM) and dividing NT2 cells (IC50=7 µM)[1]. Aloisine A functions as F508del-CFTR (temperature-corrected human airway epithelial F508del/F508del CF15 cells), G551D-CFTR (G551D-CFTR-CHO cells), and wild-type (WT)-CFTR [human airway epithelial Calu-3 and WT-CFTR-Chinese hamster ovary (CHO) cells] [2].

Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
