(R)-BI-2852 is the inactive isomer of BI-2852 and could be utilized as a control compound in experiments. BI-2852 is a KRAS inhibitor (antagonist) with nanomolar affinity based on the switch SI/II pocket designed through active molecule structure-based design. BI-2852 is mechanistically different from covalent KRASG12C inhibitors (binding switch II), binding to active KRASG12D 10 times more strongly than to KRAS wt (740 nM vs 7.5 μM). BI-2852 blocks GEF, GAP, and effector interactions with KRAS, leading to inhibition of downstream signaling and antiproliferation effects in KRAS mutant cells.
