Description Retinol (Vitamin A) is a natural vitamin that is fat soluble. Retinol plays an important role in the metabolic functions of the retina, growth and differentiation of epithelial tissues, bone growth, reproduction and immune response.
In vitro 方法:人 SH-SY5Y 神经元细胞系用 Retinol (1-20 µM) 处理 24 h,使用 MTT assay 和 SRB assay 检测细胞活力。结果:7、10 和 20 µM 的 Retinol 能显著降低细胞活力。5 µM 以下的浓度不会影响 SH-SY5Y 细胞的 MTT 还原或 SRB 掺入,但在 7 µM 浓度下,这些参数会下降,表明与对照组相比,存活细胞的数量减少了。[1]方法:人肝癌细胞 HepG2 用 Retinol (1 µM) 处理 24 h,使用 Flow cytometry 检测 Retinol 分布。结果:用 1 µM Retinol 处理 1 天后,Retinol 阳性 HepG2 细胞在总细胞群中的百分比从 17.6% 增加到 69.8%。荧光分析显示 Retinol 位于 HepG2 细胞的细胞质中。[2]
In vivo 方法:为研究胎盘药代动力学和致畸性,将 Retinol (10-100 mg/kg) 灌胃给药给妊娠第 11 天的妊娠小鼠。结果:10 mg/kg Retinol 给药后,未观察到致畸作用。药代动力学研究显示,血浆、胚胎组织、胎盘、卵黄囊膜和胚胎外液中的 retinyl esters、retinol 和 all-trans-retinoic acid 适度增加。100 mg/kg Retinol 给药后,严重胎儿畸形的发生率很高。包括胚胎在内的所有区室中的 retinyl esters 和 retinol 显著增加,极性代谢产物 all-trans-retinoic acid 和 all-trans-4-oxoretinoic acid 大量产生。[3]
