| In vivo | 斑马鱼成年雄性暴露于0.001、0.01和0.1μg/mL的Pyriproxyfen16小时。其脑组织的线粒体呼吸作用进行了评估。通过H2DCF-DA和MitoSOX估计ROS(活性氧)生成。采用Calcium Green 5 N监测钙传输。利用DAF-FM-DA估计NO(一氧化氮)合成活性。脑乙酰胆碱酯酶在体IC20为0.30μmol/LPyriproxyfen,IC50为92.5μmol/L,对zbAChE(斑马鱼乙酰胆碱酯酶)活性的抑制效应呈现竞争性。昆虫剂暴露后,复合物I/II的呼吸控制显著下降。MitoSOX测试显示O2^-(超氧阴离子)生成有Pyriproxyfen剂量依赖效应。在0.1μg/mLPyriproxyfen的浓度下,脑组织钙吸收能力损失了50%。钙释放在较低Pyriproxyfen暴露量下显示出明显的线粒体功能受损[1]。 |
