| In vitro | Lucifer Yellow CH dipotassium 工作液的配制1. 储存液的制备用 DDH2O 配制 1 mg/mL 的 Lucifer Yellow CH dipotassium。如何配置：您可以用 1 mL DDH2O 溶解 1 mg Lucifer Yellow CH dipotassium。注意事项：Lucifer Yellow CH dipotassium 储存液建议分装后于 -20℃ （短期）或 -80℃ （长期）避光保存，避免反复冻融。该产品在-20℃ 条件下可保存1个月，在-80℃ 条件下可保存半年。2. 工作液的配制用预热好的无血清细胞培养基或 PBS 稀释储存液，配制成 0.5-1% 的 Lucifer Yellow CH dipotassium工作液。注意事项：请您根据您的实验要求调整 Lucifer Yellow CH dipotassium 工作液浓度，且现用现配。细胞染色1. 细胞制备：悬浮细胞：离心后收集细胞，加入 PBS 洗涤两次，每次 5 min。贴壁细胞：去除培养基，加入胰蛋白酶消化细胞，时间为1min30s，离心弃去上清后，加入 PBS 洗涤两次，每次 5 min。2. 加入 1 mL Lucifer Yellow CH dipotassium 的工作液，室温条件下孵育 5-30 min。3. 400 g，4℃ 离心 3-4 分钟，收集细胞，弃去上清液。4. 加入 PBS 洗涤细胞两次，每次 5 min。5. 用 1 mL 无血清培养基或 PBS 重悬细胞后，在荧光显微镜或流式细胞仪下进行检测。注意事项：在加入胰蛋白酶消化细胞时，时间不宜过长，动作轻柔；PBS洗涤和重悬细胞时请勿用力使用移液枪吹打细胞。 |
