Description Gboxin is an inhibitor of oxidative phosphorylation that targets glioblastoma,inhibits the activity of F0F1 ATP synthase, with antitumour activity.
In vitro Gboxin是一种小分子化合物,专门抑制初级小鼠和人类胶质母细胞瘤细胞的生长,而不影响小鼠胚胎成纤维细胞或新生儿星形胶质细胞的生长。Gboxin能够迅速且不可逆地破坏胶质母细胞瘤细胞的氧气消耗。它依赖于其正电荷与线粒体氧化磷酸化复合物关联,此过程依赖于线粒体内膜的质子梯度,抑制F0F1 ATP合酶的活性。对Gboxin具有抵抗性的细胞需要一个功能性的线粒体透性转换孔,该孔调节pH并因此阻碍Gboxin在线粒体基质中的积累。使用代谢稳定的Gboxin类似物治疗可抑制胶质母细胞瘤异体移植物和患者源性异种移植物。Gboxin的毒性也扩展到多个器官来源的已建立的人类癌细胞系,表明癌症细胞线粒体中增加的质子梯度和pH是一种可针对抗肿瘤试剂开发的作用模式。
