| In vitro | 使用方法：1. 制备DASPEI储存液。以DMSO配制10 mM DASPEI，分装后于-20°C或-80°C避光冷藏。1.2 将储存液用预热的无血清细胞培养基或PBS稀释至5-10 μM制得工作液。根据实验需要调节浓度，临用现配。2. 细胞染色（悬浮细胞）：2.1 离心后洗涤细胞两次，细胞浓度调至1×10^6/mL。2.2 加入1 mL DASPEI工作液，常温孵育30-60分钟。2.3 400 g离心3-4分钟后去除上清，用PBS洗涤两次。2.5 重悬细胞后，应用荧光显微镜或流式细胞仪进行分析。3. 细胞染色（贴壁细胞）：3.1 细胞播种于无菌盖玻片上。3.2 移除盖玻片上多余的培养基。3.3 加入100 μL染色工作液覆盖细胞，轻轻晃动盖玻片，孵育30-60分钟。3.4 移走染色液，用培养基清洗2-3次，随后进行荧光显微镜观察。 |
