Description ADHP (10-Acetyl-3,7-dihydroxyphenoxazine) is a fluorogenic peroxidase substrate (λex=530 nm, λem=590 nm).
In vitro 为了获得Compound I的Km和kcat参数,采用了两种独立方法。首先,通过内置于荧光平板阅读器的注射器功能研究ADHP的氧化反应。自动注射器注入H2O2以触发反应,生成一系列进展曲线。对MPO介导的ADHP氧化分析显示,Km为31±4 μM,kcat为186±6/s。实验中ADHP浓度范围从1到80 μM,观测速率常数kobs随之增加;在将底物浓度固定在3到45 nM MPO的对照实验中亦是如此。从kobs对ADHP浓度作图的斜率得到的表观二级速率常数Kappon为2.1±0.2 mM/s。
