Description 4-Methylumbelliferyl oleate is a fluorescent substrate for lipases (Ex/Em=320/450 nm), which can be cleaved by lipase and can be used to measure pancreatic inhibitory activity.
In vitro 该化合物对胰脂肪酶的抑制活性是通过使用4-Methylumbelliferyl (4-MU) oleate作为底物来衡量的。反应混合物包括50 µL 0.1 mM的4-MU oleate,20 µL的McIlvane缓冲液(0.1 M柠檬酸-Na2HPO4,pH 7.4)以及5 µL的样品溶液。随后加入25 µL的猪胰脂肪酶至反应混合物中,并将总体积调整至0.1 ml。混合物在37℃下孵育10分钟后,通过在激发波长320 nm和发射波长450 nm下使用荧光多功能检测仪来测量脂肪酶释放的4-MU的量[1]。
