Description Rutin (Quercetin 3-O-rutinoside) belongs to the flavonoid group of natural products with blood-brain barrier permeability. Rutin has a wide range of biological activities including antioxidant, anti-inflammatory, anti-tumor, hypoglycemic, neuroprotective, anti-bacterial, anti-aging and so on.
In vitro 方法:不同的肿瘤细胞系用 Rutin (5-20  µM) 处理 24 h,使用 MTT assay 测定细胞活力。结果:Rutin 以浓度依赖的方式抑制不同细胞系的活力。肿瘤细胞系 SH-SY5Y、Neuro-2a、CHME、U-118、LN-229 和 SK-N-SH 的 IC50 值分别为 >40、24±4.3、15±2.6、>40、22±4.8 和 36±3.7 µM。[1]方法:人类神经母细胞瘤细胞 LAN-5 用 Rutin (25-100  µM) 处理 24 h,使用 Annexin V-FITC/PI apoptosis assay 检测细胞凋亡情况。结果:不同剂量 Rutin 的加入诱导了 LAN-5 细胞的凋亡,凋亡细胞的百分比从 4.48% 增加到 7.50%、11.26% 和 20.37%。[2]
In vivo 方法:为测试体内抗癫痫活性,将 Rutin (100-200 mg/kg) 腹腔注射给 BALB/c 小鼠,每天一次,持续七天。最后一天注射 kainic acid (10 mg/kg) 诱导癫痫。结果:Rutin 组癫痫发作评分明显低于对照组。Kainic acid 给药后第 1 天和第 21 天,Rutin 组海马丙二醛水平显著低于对照组。Rutin 组在 kainic acid 给药后第 1 天观察到的硫醇水平高于对照组。[3]
