Description Marein shows neuroprotective effect on PC12 cell damage induced by methylglyoxal, which is due to a reduction of damage to mitochondria function and activation of the AMPK signal pathway, it may be a potent compound for preventing/counteracting diabetic encephalopathy.
In vitro 为了明确来自降血糖植物黄花菜(Coreopsis tinctoria)的主要化合物Marein是否能防止MG诱导的PC12细胞损伤,我们在含有MG和Marein的培养基中培养PC12细胞。研究发现,Marein减缓了MG引起的以下变化:线粒体膜电位(ΔΨm)、线粒体透性转换孔(mPTPs)、细胞内Ca2+水平、活性氧种(ROS)的产生、谷胱甘肽(GSH)/氧化型谷胱甘肽(GSSG)和腺苷三磷酸(ATP)水平,以及凋亡细胞百分比的增加。Marein还增加了乙醛缩酶I (Glo1)活性、磷酸化AMPKα(Thr172)和Bcl-2表达,并减少了Bax、caspase-3和caspase激活的核酸酶(ICAD)的激活。值得注意的是,Marein预处理减弱了复合体C对p-AMPK的失活作用。分子对接模拟显示,Marein与AMPK的γ亚基发生了相互作用[1]。
