Description Lupiwighteone has anti-angiogenesis potential, it also has anticancer and cancer preventive effects on SH-SY5Y cells.
In vitro 本研究旨在检验Lupiwighteone的抗肿瘤活性,探究其在人类前列腺癌细胞系(DU-145)中的抗肿瘤机制,并评估其在人脐静脉内皮细胞系(HUVEC)中的抗血管生成活性。结果表明,Lupiwighteone通过3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴化物(简称MTT)检测,能够以浓度依赖和时间依赖的方式抑制DU-145和HUVEC细胞的增长。流式细胞术分析显示,Lupiwighteone能诱导DU-145细胞周期阻滞、细胞凋亡、线粒体膜电位丧失和细胞内活性氧的增加。Western blot分析发现,Lupiwighteone处理后,观察到Bax、细胞色素c、caspase-3和PARP-1蛋白表达上调,而Bcl-2、procaspase-9和p-Akt蛋白表达下调。此外,还研究了Lupiwighteone对HUVEC细胞行为的影响。
