Description Gelsevirine has anxiolytic activity, may be via the agonist action of glycine receptor in the brain.
In vitro Four alkaloid compounds were assessed for cytotoxic activity using the microculture 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay for cellular viability. The results showed that Koumine,Gelsemine,Gelsenicine and Gelsevirine could significantly inhibite the proliferation of human carcinoma cell line SW480 and MGC80-3 in a dose-dependent manner. The 50% inhibiting concentration(IC50)of Koumine,Gelsemine,Gelsenicine and Gelsevirine on SW480 cells were 0.45±0.10,0.76±0.28,0.52±0.22 and 1.41±0.06 mmol·L-1 respectively, and were 0.82±0.19,1.20±0.33,1.14±0.23 and1.22±0.11 mmol·L-1 respectively on MGC80-3 cells.Koumine,Gelsemine and Gelsenicine also could inhibite the proliferation of TE-11 and HepG2 cells in a dose-dependent manner.IC50 of Koumine,Gelsemine and Gelsenicine on TE-11 cells were 0.74±0.05,1.94±0.30 and 1.73±0.35 mmol·L-1 respectively, and were 1.26±0.32,1.82±0.35 and 1.79±0.54 mmol·L-1 respectively on HepG2 cells.
