Description D-Galactose (Alpha-D-galactose) is a naturally occurring hexose sugar that combines with glucose to form lactose. D-Galactose is often used in animal aging models.
In vitro 方法:星形细胞 CRT 和大鼠原代星形胶质细胞用 D-Galactose (10-100 g/L) 处理 72 h,使用 WST-1 assay 检测细胞活力。结果:40 g/L 以上的 D-Galactose 以剂量依赖性方式显著抑制细胞活力。[1]方法:胶质母细胞瘤细胞系 C6 用 D-Galactose (28-222 mM) 处理 8 天,使用 Western Blot 检测靶点蛋白表达水平。结果:与对照组相比,D-Galactose 处理组中裂解的 cleaved-caspase-3/caspase-3 水平显著降低。[2]
In vivo 方法:为研究对动物行为的影响,将 D-Galactose (50-200 mg/kg) 皮下注射给 C57 小鼠构建衰老模型,每天一次,持续八周。结果:D-Galactose 可在 50-100 mg/kg 范围内以剂量依赖性方式诱导 C57 小鼠的行为损伤,高于 100 mg/kg 的剂量不会进一步恶化其行为。[3]
