Description Cerulenin is a natural inhibitor of fatty acid synthase (FAS). It is produced by the fungus Cephalosporium caeruleus.
In vitro Cerulenin与StAR过表达具有相似效应。StAR通过减少主动脉内皮细胞中的脂质合成和摄取,以及减轻PA引起的炎症和一氧化氮(NO)生物可用性降低,从而缓解了由PA引发的内皮细胞功能障碍[1]。
In vivo 接受了cerulenin治疗的ob/ob肝脏的组织学剖析展现了脂质含量的显著降低,这种降低既与剂量相关又与时间相关。60 mg/kg cerulenin进行七天治疗,无论是每天还是隔天给药,均展示了最大幅度的脂肪染色减少,在肝脏的某些区域,ORO染色甚至完全消失(图3D-F)。60 mg/kg cerulenin的两天治疗结果显示脂肪含量有适度减少。相比之下,30 mg/kg的cerulenin的效果不如60 mg/kg剂量那样显著,但对于脂肪含量的影响仍然明显,除了两天治疗的情况(数据未示)[2]。
