Description Bafilomycin A1 belongs to the macrolide class of antibiotics and is a V-ATPase inhibitor (IC50=0.44 nM) that is specific and reversible. Bafilomycin A1 is an inhibitor of the late phase of autophagy, blocking the fusion of autophagosomes with lysosomes. Bafilomycin A1 also induces apoptosis.
In vitro 方法:人胰腺癌细胞 Capan-1 用 Bafilomycin A1 (1-100 nM) 处理 72 h,使用 MTT 方法检测细胞生长抑制情况。结果:Bafilomycin A1 剂量依赖抑制 Capan-1 细胞生长,IC50 为 5 nM。[1]方法:人骨肉瘤细胞 MG63 用 Bafilomycin A1 (1 μmol/L) 处理 6-24 h,使用 Western Blot 方法检测靶点蛋白表达水平。结果:Bafilomycin A1 诱导细胞凋亡相关蛋白 p53 和 Beclin 1 水平显著增加,诱导自噬相关蛋白 p62 和 LC3-I 的表达水平显著降低,而 LC3-II 的水平升高。[2]方法:人肝癌细胞 BEL7402 和 HepG2 用 Bafilomycin A1 (5  nM) 孵育 24 h,使用 Flow Cytometry 方法检测细胞周期情况。结果:Bafilomycin A1 增加了 BEL7402 和 HepG2 细胞在细胞周期 G1 期的百分比,同时 G2/M 期的细胞减少,表明 G1 期停滞。[3]
In vivo 方法:为检测体内抗肿瘤活性,将 Bafilomycin A1 (0.1-1 mg/kg) 腹腔注射给携带人急性淋巴性白血病肿瘤 B-ALL 的 NOD-SCID 小鼠,每天一次,持续三天。结果:Bafilomycin A1 通过靶向白血病细胞延长异种移植物小鼠的生存期并改善其病理学。[4]方法:为研究自噬在慢性伤口愈合中的作用,将 Bafilomycin A1 (1 mg/kg) 单次腹腔注射给糖尿病 db/db 小鼠。结果:Bafilomycin A1 治疗显著加速了 db/db 小鼠的伤口愈合,并发挥了良好的愈合效果。Bafilomycin A1 可能通过促进细胞增殖、胶原生成和调节炎症平衡来加速糖尿病慢性难治性伤口的愈合。[5]
