Bioactivity Mezigdomide (CC-92480), a cereblon E3 ubiquitin ligase modulating drug (CELMoD), acts as a molecular glue. Mezigdomide shows high affinity to cereblon, resulting in potent antimyeloma activity[1].
Invitro Mezigdomide is the second cereblon modulator. Mezigdomide-induced loss of Aiolos and Ikaros in cultures of PBMCs resulted in the activation of T cells and increased production of IL-2 and IFN-γ. Mezigdomide is effective in CC-5013, CC-4047, and CC-220-resistant cell lines. It exerts single-agent induction of apoptosis and exhibits remarkable synergy with NSC 34521[1].
