Description MMP-1 Substrate II is a collagenase substrate with the sequence of Dnp-Pro-Leu-Gly-Cys(Me)-His-Ala-D-Arg-NH2. In an approach to optimize substrate specificity, two amino acids have been substituted at the cleavage site of the collagenase substrate, Dnp-Pro-Leu-Gly-Leu-Trp-Ala-D-Arg-NH2. MMP-1 Substrate II possesses a 10-fold better kcat/Km value for human fibroblast collagenase than the FRET substrate Dnp-Pro-Leu-Gly-Leu-Trp-Ala-D-Arg-NH2. However, monitoring of the enzyme activity using MMP-1 Substrate II requires HPLC analysis as the quencher (Trp) has been removed by the amino acid substitutions.
