Description H-Glu-Ala-Gly-Asp-Asp-Ile-Val-Pro-Cys-Ser-Met-Ser-Tyr-Thr-Trp-Thr-Gly-Ala-OH also called S5A/5B, EAGDDIVPCSMSYTWTGA, which mimics the NS5A-5B cleavage junction of the nonstructural protein (NS) is an efficient substrate used by Shimuzu et al. to detect NS3 protease activity in a standard assay (Km = 411 µM and kcat = 0.14 min-1, without NS4 activator). Cleavage occurs between Cys and Ser. The cleavage products are analyzed by RP-HPLC.
